目的

初步探索健康成年人唾液中胃蛋白酶含量范围,为临床检测、诊断咽喉反流性疾病提供参考依据。

方法

2016年9月1日~9月30日在海军总医院耳鼻咽喉头颈外科门诊招募50名健康成年人作为受试者,收取受试者晨起空腹情况下黏白唾液,封存于1.5 ml无菌EP管内,并在30 min内将EP管放置于-28 ℃冰箱中冷冻,最长冷冻时间不超过2周;采用人胃蛋白酶ELISA检测试剂盒测定受试者唾液中胃蛋白酶含量。

结果

50名健康受试者晨起痰液中胃蛋白酶含量为(24.22±6.94)ng/ml,95% CI为(22.18,26.06)ng/ml。其中,男性受试者唾液中胃蛋白酶含量为(22.78±1.35)ng/ml,95% CI为(20.00,25.57)ng/ml;女性受试者唾液中胃蛋白酶含量为(25.67±1.39)ng/ml,95% CI为(22.80,28.54)ng/ml。

结论

健康成年人唾液中存在一定含量的胃蛋白酶,检测唾液胃蛋白酶含量有望用于咽喉反流性疾病的诊断。

Objective

To measure the pepsin level in saliva of the healthy adults in order to find a method to diagnose the laryngopharyngeal reflux disease by measurement of the pepsin level in the saliva of patients.

Methods

From Sept.1 2016 to Sept.30, 2016 in Department of Otolaryngology Head Neck Surgery, Navy General Hospital, 50 healthy adults were randomly recruited as subjects, and the saliva was collected into 1.5ml sterile EP tubes after awaking in the morning, and the tubes were placed in a refrigerator of -28 ℃ within half an hour, meanwhile the longest freezing time was less than 2 weeks.The level of pepsin in saliva was measured by human pepsin ELISA kit.

Results

Among the 50 subjects, the average content of pepsin in saliva was(24.22±6.94)ng/ml and the 95% confidence interval was(22.18, 26.06)ng/ml.The average content of pepsin in saliva was(22.78±1.35)ng / ml and 95% confidence interval was 20.00, 25.57)ng/ml in male group.The average content of pepsin in saliva was(25.67±1.39)ng/ml and 95% confidence interval was(22.80, 28.54)ng/ml in female group.

Conclusion

The pepsin can be detected in the saliva of the healthy adults, the level of the pepsin in the saliva of the patients may be used to the diagnosis of the laryngopharyngeal reflux disease in the future.
