The role of IL-6 and PD-L1 in occurrence of Gastroesophageal reflux disease

Abstract:

Objective

To investigate the role of Interleukin-6 (IL-6) and Programmed Death Ligand 1 (PD-L1) in the pathogenesis of Gastroesophageal reflux disease (GERD).

Methods

In vitro, culture rat esophageal epithelial cells and CD4+T cells and establish a coculture system with normal rat esophageal epithelial cells and CD4+T cells. In the experimental group, chenodeoxycholic acid, pancreatin, and hydrochloric acid were added to the culture medium in order to adjust the pH of the medium to 4, 5, and 6, respectively; cells were cultured for 12 h, 24 h, and 48 h. The control group cells were cultured normally without any stimulation. ELISA and Western blot were used to detect the expression levels of IL-6 and PD-L1 in each group. The experimental data is expressed as

±s. The overall comparison uses repeated measures analysis of variance, and no correction is required to meet the sphericity test If correction is required, Green house-Geisser correction is used.

Results

With the increase of pH concentration and the extension of culture time, the expression levels of IL-6 and PD-L1 in each group were significantly increased (P<0.05); expression levels of IL-6 and PD-L1 in epithelial cells and CD4+ cell co-culture system were reached the highest level (P<0.05).

Conclusion

The inflammatory cytokine IL-6 may promote the expression of PD-L1 in GERD and play an important role in the occurrence and development of GERD.
